1、姜黄素对卵巢癌微血管内皮细胞生物学特性及乙酰肝素酶的影响杨鹰,谢荣凯,阎萍,韩静第三军医大学附属新桥医院妇产科重庆400038【提摘要】:目的探讨姜黄素对卵巢癌微血管内皮细胞体外生物学特性及乙酰肝素酶表达的影响。方法体外培养卵巢癌微血管内皮细胞,分为正常对照组(正常组)和姜黄素组(浓度分别为2μg/ml、10μg/ml、50μg/ml和、250μg/ml),观察时间为加入姜黄素共孵育后24h、48h和、72h。四甲基偶氮噻唑蓝法(MTT法检测细胞增殖抑制率,流式细胞仪检测细胞周期变化,免疫荧光法和RT-PCR法检测内皮细胞乙酰肝素酶表达,生化法检测细胞上清液丙
2、二醛(MDA)含量及乳酸脱氢酶(LDH)活力。结果姜黄素组细胞生长抑制率增加,有显著的剂量依赖效应。姜黄素组增殖指数显著下降,乙酰肝素酶mRNA水平和蛋白水平显著下降,细胞培养上清液MDA水平及LDH活力显著升高。结论姜黄素(2~250μg/ml)能显著抑制人卵巢癌微血管内皮细胞增殖活性,降低乙酰肝素酶mRNA转录和蛋白表达。关键词】:姜黄素;卵巢癌;血管内皮细胞;乙酰肝素酶【中图分类号】:R737.31文献标识码:AEffectofcurcumineonthebionomicsandheparanaseexpressionofbloodvesselendot
