3、high-yieldcrops.ofyield.Here,wereportthecloningandfunctionalanalysisWecarriedouthigh-resolutionmappingwithhomozygousrecom-ofTHOUSAND-GRAINWEIGHT6(TGW6),agenefromthebinantplantsanddelimitedtheTGW6-containingregionto4.9kbIndianlandracericeKasalath.TGW6encodesanovelprotei

4、nbetweenthemarkersG214andG232.Inthisgenomicregion,therewithindole-3-aceticacid(IAA)-glucosehydrolaseactivity.wasonlyoneORF,Os06g0623700,predictedbytheRiceAnnotationInsinkorgans,theNipponbaretgw6alleleaffectsthetimingProject(RAP-DB;seeURLs)(Fig.1a).Thisputativegene,avia

5、bleofthetransitionfromthesyncytialtothecellularphasebycandidateforTGW6,hasonlyoneexon.TheKasalathallelecontainedcontrollingIAAsupplyandlimitingcellnumberandgrainsixnucleotidesubstitutionsanda1-bpdeletionatnucleotide313length.Mostnotably,lossoffunctionoftheKasalathallel

6、e(Fig.1b)comparedtotheNipponbareallele.The1-bpdeletioncausedenhancesgrainweightthroughpleiotropiceffectsonsourceaframeshiftandpreventedtheproductionofthematureprotein,organsandleadstosignificantyieldincreases.Ourfindingsowingtotheintroductionofaprematurestopcodon(Fig.1

7、bandsuggestthatTGW6maybeusefulforfurtherimprovementsinSupplementaryFig.1),whereasthesixnucleotidesubstitutionswereyieldcharacteristicsinmostcultivars.notexpectedtoaffecttheprotein.Therefore,theframeshiftmighthavegivenrisetofunctionallossoftheNipponbare-typeTGW6pro-Duri
