6、······························50个人简历·······································································································51新型复合支架对骨髓间充质干细胞增殖及分化影响的研究摘要目的:创伤、退行性病变或肿瘤等原因导致的骨缺损仍然是骨骼修复重建的主要难题。同种异体骨移植和自体骨移植是当前治疗骨缺损的最佳手段。而这两种治疗都有其局限性,如供体组织有限、供体部位发病率高、手术复杂、同种异体移植物疾

7、病传播或免疫排斥的可能性、且移植物无法准确塑形替代原组织的生物力学特性。如何实现骨缺损修复重建一直是当今医学界的难题,组织工程支架为解决这一难题提供了很好的思路。本实验旨在采用冷冻干燥法制备胶原蛋白/壳聚糖/羟基磷灰石三维仿生骨组织工程支架,并观察胶原蛋白/壳聚糖/羟基磷灰石三维复合支架对骨髓间充质干细胞的增殖和分化的影响。目的是研究组织工程支架治疗骨缺损修复重建的可行性。方法:1.大鼠骨髓间充质干细胞(BMSCs)的分离培养和分化鉴定:抽取清洁级健康雄性SD大鼠骨髓,密度梯度离心法提取单个核细胞,贴壁法分离骨髓间充质干细胞。通过CCK-8法检测

8、细胞生长曲线;成骨诱导分化培养后,进行茜素红染色检测矿物结节染色评估成骨分化情况。2.胶原/壳聚糖/羟基磷灰石复合支架制备:采用冷冻干燥
