1、捅要脂肪酸是维持人体正常生理功能的必须物质,分为饱和脂肪酸、单不饱和脂肪酸和多不饱和脂肪酸。n.3多不饱和脂肪酸是一类备受医学界关注的多不饱和脂肪酸,已证明它的摄入具有治疗肥胖和防治代谢类疾病的作用。本实验将30只5周龄健康大鼠分为n.6多不饱和脂肪酸饮食组、n.3多不饱和脂肪酸饮食组和对照组,自由摄食饮水8周,记录每周大鼠进食量、体重和体长,实验结束时处死大鼠,取其血清、胃、脂肪、肝脏、盲肠及盲肠内的粪便和附睾脂肪。设计引物,用qRT-PCR检测双歧杆菌(Bifidobacterium,Bif)、乳酸杆菌(Lactobacillus,
2、Lac)、Akkermansiamuciniphila(Akk)、艰难梭状芽孢杆菌(Clostridriumdifficile,Clo)、金黄色葡萄球菌(Staphylococcusaureus,Sta)、大肠杆菌(E.coli,Eco)和脂肪酸合成酶(fattyacidsynthetase,FAS)、脂联素(adiponectin)、nesfatin.1和ghrelin的表达水平,用免疫印迹法检测nesfatin.1蛋白的表达水平,并将大鼠盲肠进行石蜡包埋、切片、HE染色,测量其粘膜厚度,来研究多不饱和脂肪酸饮食对大鼠肥胖程度、肠道菌
