4、schangesinthefunctionofastrocytegapjunctionoccurringintheratprefrontalcortex(PFC)afterchronicunpredictablestress(CUS),arodentmodelofdepression.AnimalsexposedtoCUSandshowingbehavioraldencitsinsucrosepreferencetest(SPT)andnoveltysuppressedfeedingtest(NSFT)exhibi
5、tedsignificantdecreasesindiffusionofgapjunctionchannel-permeabledyeandexpressionofconnexin43(Cx43),amajorcomponentofastrocytegapjunction,andabnormalgapjunctionalultrastructureinthePFCFurthermore,weanalyzedtheeffectsoftypicalantidepressantsfluoxetineandduloxeti
6、neandglucocorticoidreceptor(GR)antagonistmifepristoneonCUS-inducedgapjunclionaldysfunctionanddepressive-likebehaviors.ThecellularandbehavioralalterationsinducedbyCUSwerereversedand/orblockedbytreatmentwithtypicalantidepressantsormifepristone,indicatingthatthem
7、echanismoftheirantidepressantactionmayinvolvetheameliorationofgapjunctiondysfunctionandthecellularchangesmayberelatedtoGRactivation.WetheninvestigatedtheeffectsofpharmacologicalgapjunctionblockadeinthePFCondepressive-likebehaviors.Theresultsdemonstratethatcarb
8、enoxolone(CBX)infusionsinducedanhedoniainSPT,andanxietyinNSFT,andCx43mimeticpeptiaesGap27andGap26alsoinducedanhedonia,acoresymptomofdepression.Together,thisstudysupportsthehypothes
