2、turedonthenanostructuredcoatingsurfaceswereobservedinvitro.24Tiimplantswithgradientcoatingswereplantedinfemoralcondylesof12NewZealandrabbitsrandomlyandtheuntreatedTiimplantswithsamesizewereplantedasthecontrols.At1,3and6monthsafterimplantation,theanimalsweresacrificedafterx-rayexamina

3、tionandthetissuearoundtheimplantsweretakenoutforprepareofhardtissuesectionandgroundsection.Quantityofnewbonewasmeasuredbytetracyclinefluorescencestaining.VonGiesonstainedmicrographswereusedtoobservethehealing,bonedepositionandosteointegrationattheinterfacebetweenboneandimplant.2.Prep

4、arationofnanostructuredtitaniumsurfacebyplasticdeformationandobservationofthesurfacepropertiesandthebiocompatibilityMedicalTisheets(10mm×10mm×1mm)weresubjectedtoplasticdeformationunderthecertainconditionfordifferenttime.Theplasticdeformationmaterialsweredividedintothreegroups:groupⅠ(

5、30minutes,n=6),groupⅡ(60minutes,n=6)andgroupⅢ(90minutes,n=6),accordingtothetimeofpreparation.Thecharacteristicsof上海交通大学医学院2005级硕士学位论文plasticdeformationmaterialwereevaluatedbySEM,TEM,XRD,CLSMandDMA.Theuntreatedtitaniumwasusedascontrolgroup.Saos-2celllinewasculturedonthedifferenttitani

6、umsurfaces.Thefeaturesoftitaniumsurfaceandtheeffectsofnanostructuredtitaniumsurfacesoncelladherence,proliferationandshapewereexaminedusingfluorescencemicroscope,LSCMandMTTtesting.RT-PCRwasusedtoassessthealterationofBMP-4geneexpression.ANOVAwasperformedusingSAS6.0softwarepackage.RESUL

7、T:1.Thegradientcoatingswereporousanditssurfacewerecomposedofirregularrod-likenano-HAcrystalswiththediameterssmallerthan100nm.TheinterfacebetweenthecoatingsandTiwasdenseandnocrackswerefound.Thebondstrengthwas35.5±3.5MPabetweencoatingandTi.CellularadherenceandproliferationofSaos-2cells
