2、engineering,oilextraction,textileindustry,wastewatertreatment,bio-energyproduction,etc.Ithasmanyresearchpapersaboutcellulose,buttheseriousproblemwasthestrainsushasTrichodermareesei,Penicilliumwhichproducedcellulosehadlowertheactivityofcellulase.Sotheamoutofthecellulasewerelagerly,whichcausedhi

3、ghcost.High—yieldcellulaseproducingstrain,theoptimumconditionsoffermentationandsuitableconditionofenzymereactionsystemcaneffectivelyreducecellulaseexpenses.Inthisstudy,anexcellentcellulase-producingstrainWasscreenedfromtheforestfloorunderrottingwoodandsoilinYueluMountain.Itwasmutatedsequential

4、lybydifferentphysicalmethods,andmicroscopicobservationand16srDNAsequencealignmentwereusedforidentificationmutantstrain,besides,thefermentationconditionandtheconditionsenzymereactionsystemandenzymaticpropertieswerestudied.Themainresultswereasfol!ows:Anexcellentcellulase—producingstrainA24wasscr

5、eenedfromtheforestfloorunderrottingwoodandsoilinnature,andtheactivityofCMCaseandFPAasewere20.37U/mL,1.20U/mL,respectively.TheoriginalstrainA24WastreatedwithUVmutagenesis,andthebestUVmutationtimewas7min.Inthiscondition,amutantstrainUV-37hadgoodgeneticstabilityandtheactivityofCMCaseandFPAasewere

6、25.35U/mLand1.33U/mL,respectivly.Thebest60Co吖raymutationdosewas0.6KGyforthemutantstrainUV-37.Inthiscondition,amutantstrainCo-07whohadgoodgeneticstabilityandtheactivityofCMCaseandFPAasewerehigherthantheoriginalstrain137.16%and118.80%.Themorphologicalcharacteristicsofcolonyandmyceliumandsporewir
