2、hmarrowmesenchymalstemcells(MSCs),comparedtheeffectoffemoraldefectrepair,preliminarystudyfortreatmentofbonedefectswaysoftissueengineering.[Methods]:Animalmodelsoffemoralbonedefectwereestablishedin30Sprague—Dawleyrats.Tlleyweredividedinto3groupsrandomly:Ing

3、roupA,defectwasfilledwithnone;ingroupBwithnHAC—MSCscompound;ingroupCwithMSCs/Shh—nHACcompound.MSCswereisolated,purified,cultured,identifiedandobservedcontinually.Scaffoldmaterialswerecutintoblockswiththesizeo勋6mmx5mmx5mm.Cellsofthe3rdgenerationswereCO—cult

4、uredwithMSCs/Shh-nHACinvitro.Thecompositegraftsweretestedbyscanningelectronmicroscope.Abonedefect(5mminlength)wascreatedattwofemoralineachmouse.Thecompositegraftswereimplantedintosegmentalfemoraldefectsinratsthroughopenoperation.Thecurativeeffectwasevaluat

5、edbygeneralobservation,radiographicexamination,histologicanalysisat6and12weeks.[Results】:After14days’compositecultivationoutsides,MSCsexhibitednewcellgrowthbyscanningelectronmicroscope.AtSixweeksaftertheoperation,whilegroupAshowedobviousbonedefects,whichre

6、mainedtobeseenafteranother6weeks,groupBshowedcloudynewbomosteotylus,whichgrewintogreatquantityafterIII江苏大学硕士学位论文another6weeks,butstillnotinbone-mouldingprocess,andgroupCshowedthedegradationoftheShh—nHACscaffoldandafewnewboneformationaswellasthecloudbetween

7、bonedefectsbyradiographicexamination,whichgrewintocontinuousosteotylUSandsmoothofbonemarrowcavityandbonetrabeculainsteadoftheplantedMSCs/Shh-nHACcompound.Thenewboneformationwassignificantlydifferentbetweentheexperimentalandthecontrolgroupsbyradiographicexa

8、minationat6and12weeksafteroperation(P<0.05).Andthespeedandquantityofnewboneformationwasalsosignificantlydifferentbetweentheexperimentalandthecontrolgroupsbyhistologicanalysis(P<0.05).[Conclusion]:MSCsareideal
