3、umors.Thehallmarksconstituteanorganizingprincipleforrationalizingthecomplexitiesofneoplasticdisease.Theyincludesustainingproliferativesignaling,evadinggrowthsuppressors,resistingcelldeath,enablingreplicativeimmortality,inducingangiogenesis,andacti-vatinginvasionandmetastasis.Underlyingthese
4、hallmarksaregenomeinstability,whichgeneratesthegeneticdiversitythatexpeditestheiracquisition,andinflammation,whichfostersmultiplehall-markfunctions.Conceptualprogressinthelastdecadehasaddedtwoemerginghallmarksofpotentialgeneralitytothislist—reprogrammingofenergymetabolismandevadingimmunedest
5、ruction.Inadditiontocancercells,tumorsexhibitanotherdimensionofcomplexity:theycontainarepertoireofrecruited,ostensiblynormalcellsthatcontributetotheacquisitionofhall-marktraitsbycreatingthe‘‘tumormicroenvironment.’’Recognitionofthewidespreadapplicabilityoftheseconceptswillincreasinglyaffect
6、thedevelopmentofnewmeanstotreathumancancer.INTRODUCTIONvatedbythesedevelopments,wenowrevisittheoriginalhall-marks,considernewonesthatmightbeincludedinthisroster,Wehaveproposedthatsixhallmarksofcancertogetherconsti-andexpanduponthefunctionalrolesandcontributionsmadetuteanorganizingprinciplet
