5、··37个人简历·············································································38中文摘要苦苣菜水提物的抗炎作用及其作用机制研究摘要苦苣菜(学名:SonchusoleraceusL.),又名:苦菜、滇苦菜、苦荬菜、拒马菜、苦苦菜、野芥子,英名CommonSowthistle,属于菊科(Compositae),苦苣菜属(Sonchus)l~2年生的草本植物。生长分布几乎遍及全球,主要生于海拔170~3200米左右的山坡或山谷林缘、林下或平地

6、田间、空旷处或近水处。在我国分布广泛,辽宁、河北、陕西、新疆、四川等地都可见到。苦苣菜属植物的主要活性成分是倍半萜类和黄酮类,此外,还有香豆素类、甘油酸酯苷类、木脂素类等。该属药用植物及其提取物具有多方面药理作用,如保肝、利尿、降压、抗凝血、抗炎、抗菌、抗肿瘤等,同时含有维生素C、维生素B1、胡萝卜素等多种维生素,和铜、镁、铁、锌等多种微量元素。具有营养高、产量高、适口性好等优点,有很高的营养和药用价值,开发前景十分广阔。特别是近年来,许多实验研究表明该属植物具有抗菌、抗炎作用,临床上也常用于治疗胃炎、肠炎、肺热咳嗽、咽喉肿痛等

7、多种炎症性疾病。但具体的机制研究报道较少,且涉及抗氧化、减少NO生成、抑制凋亡等多个方面,缺乏梳理和深入阐明。目的:通过二甲苯致小鼠耳廓肿胀和角叉菜胶诱导小鼠后足跖肿胀建立急性炎症模型,研究苦苣菜水提物对多种急性炎症的拮抗作用,并通过检测小鼠炎性组织液和血清中COX-2、p38MAPK、TNF-α、IL-6等多种炎症相关因子的含量变化,进一步探讨苦苣菜水提物对这些细胞因子、炎性介质的影响,对机体炎症相关通路的调节作用,初步分析苦苣菜水提物抗炎免疫活性的有关机制。方法:给小鼠耳廓内外涂抹二甲苯,引起炎症介质释放,造成耳廓急性渗出性

8、炎性水肿。给小鼠足跖皮下注射角叉菜胶,诱导形成足跖肿胀。-1造模前每组小鼠分别灌胃给予苦苣菜水提物(0.26g•10g、0.13g•10g-1-1、0.065g•10g)、等量生理盐水,连续5天。末次给药30分钟后制备耳廓肿胀模型,致炎30分钟后,脱颈椎处死小鼠
