7、···49中文摘要低剂量节拍化疗对卵巢癌细胞SKOV3血管生成拟态的影响及其机制的探讨摘要目的:血管生成拟态(vasculogenicmimicry,VM)是高侵袭性恶性肿瘤细胞所具有的自我血供的微循环结构。本实验通过对荷人卵巢上皮性癌SKOV3细胞裸鼠异种移植瘤模型行不同模式的顺铂(DDP)化疗获取移植瘤原代细胞,比较接受低剂量节拍化疗(low-dosemetronomicchemotherapy,LDM)和最大可耐受剂量化疗(maximumtolerateddosechemotherapy,MTD)后SKOV

8、3移植瘤原代细胞形成VM的能力、增殖力及侵袭力,同时分析不同化疗模式对干细胞标记物CD133、ALDH1蛋++--白表达的影响,并比较CD133ALDH1和CD133ALDH1细胞形成血管生成拟态的差异,以探讨不同模式化疗对卵巢癌SKOV3细胞形成VM的影响及其可能机制。方法:71建立裸鼠移植瘤模型并分组化疗:体外培养SKOV3细胞,将1×10个细胞接种于雌性裸鼠右侧的大
