2、tractBackground:TheproductivityofthemedicinallysignificantperennialherbRehmanniaglutinosaisseverelyaffectedafterthefirstyearofcropping.Whilethereissomeinformationavailabledescribingthephysiologicalandenvironmentalcausesofthisyielddecline,thereisasyetnodatareg

3、ardingthechangesingeneexpressionwhichoccurwhenthespeciesiscontinuouslycropped.Results:Usingamassivelyparallel(Solexa)DNAsequencingplatform,itwaspossibletoidentifyandquantifytheabundanceofalargenumberofR.glutinosamiRNAs.WecontrastedthemiRNAcontentoffirstyearcr

4、opplantswiththatofsecondyearcropones,andwereabletoshowthatof89conserved(belongingto25families)andsixnovelmiRNAs(sixfamilies),29oftheformerandthreeofthelatterweredifferentiallyexpressed.ThethreenovelmiRNAswerepredictedtotargetsevengenes,andthe29conservedones30

5、8genes.Thepotentialtargetsof32ofthesedifferentiallyexpressedmiRNAsinvolvedinthemaintranscriptionregulation,plantdevelopmentandsignaltransduction.AfunctionalanalysisofthedifferentiallyexpressedmiRNAssuggestedthatseveraloftheproposedtargetscouldbedirectlyorindi

6、rectlyresponsibleforthedevelopmentofthetuberousroot.Conclusion:WehavecompareddifferentialmiRNAsexpressioninthefirstyearcrop(FP)R.glutinosaplantsandsecondyearcrop(SP)ones.Theoutcomeidentifiessomepotentialleadsforunderstandingthemolecularbasisoftheprocessesunde

7、rlyingthedifficultyofmaintainingtheproductivityofcontinuouslycroppedR.glutinosa.BackgroundcropoverseveralseasonshavefailedtoovercometheRehmanniaglutinosaL.isaperennialherbaceousspeciesmajordeclineinproductivity,asthetubersareincreas-belongingtotheScrophularia
