2、omplementaryDNA(cDNA)preparedagainsttheA59nucleocapsidproteinmessengerRNA,wehaveinvestigatedtheextentofhomologybetweenA59andfourotherstrainsofmurinehepatitisvirus(MHV).AnalysisbyhybridizationkineticsoftheannealingbetweenA59[S2P]cDNAandinfectedcellRNAfromtheoth

3、erfourMHVstrainsdemonstrated7040%homology.Bygeltransferanalysis,theA59[S2P]eDNAwasabletodetectsubgenomic-sizevirus-specificRNAsincellsinfectedwithallofthefiveMHVstrains.AsimilarpatternofsevenviralRNAswasdetectedincellsinfectedwithA59,MHV-1,MHV-3,andJHM.Incontr

4、ast,cellsinfectedwithMHV-Scontainedsevenvirus-specificRNAs,ofwhichonlythetwosmallestspeciescomigratedwithRNAsfromtheotherfourstrains.TheresultssuggestthataspreviouslyshownwithA59(S.Cheley,R.Anderson,M.J.Cupples,E.C.M.LeeChan,andV.L.Morris(1981)Virdogy,112,596-

5、604),allMHVstrainstestedencodeanestedsetofsubgenomicRNAs.Analysisof[?l]methionine-labeledviralproteinsbySDS-polyacrylamidegelelectrophoresisre-vealedthateachstrainofMHVspecifiedfourmajorviralpolypeptideswithapparentmolecularweightsverysimilartothosepreviouslyr

6、eportedfortheE2,N,El,andPElpolypeptidesofA59.ThestrongdegreeofinterstrainhomologyamongthefiveMHVstrainsinvestigatedwasconfirmedbycomparativechymotrypticpeptidemappingoftheviralNproteins.Amajorityofthechymotrypticpeptidesfromeachofthe[35S$nethionine-labeledNpro
