2、,Ottawa,Ontario,CanadaWith5FiguresAcceptedMay8,1981SummaryApersistentinfectionbyhumancoronavirus229E(HCV/229E)wasestab-lishedinahumancontinuouscellline(L132).FollowingtheinitialinfectionwithstockHCV/229E,severalcultureswereestablishedofwhichtwo(HVtandIIV4)havebeenmaintainedbycontinouspassagefortwo
3、years.Theseeultureshaveshedhightitresofinfectiousviruscontinuouslyintothesupernatantfluidsincetheirinitiation.Thepersistentlyinfectedceilswereresistanttohomologoussuper-infectionbutsupportedpoliovirusreplicationtonormaltitres.Preliminarytestsindicatedthat50--100percentofthecellscontainvirus.Neithe
4、rinterferonnorreversetranseriptasecouldbedetectedintheseculturesandthepresenceofdefectiveinterferingparticlescouldnotbedemonstrated.VHtandVH4corona-viruses,isolatedfromthesepersistentlyinfectedcultures(HV)andidentifiedby229Eantiserumneutralization,weremoreeytoeidalthantheparentvirusasjudgedbyplaqu
5、echaracteristicsandCPEhowever,theywereindistinguishableonthebasisofdensity,EMmorphology,a.ndgenomesize.Presentevidenceindicatesthattemperatureplaysanimportantbutasyetundeterminedroleintheestablishmentandmaintenanceofstable229Epersistentlyinfectedcellcultures.IntroductionCoronavirusesareimportantpa
6、thogensofhumansandanimalscausingavarietyofrespiratoryandentericinfectionsofworldwidedistribution(17).Inadditiontothewellknownacuteinfectionsofman,swine,dogs,eats,calvesandchickens,longtermeoronaviralinfectionshavebeendescribedinchickens,miceandswine.Also,ithasbeenreportedthathumaneoronavirus(HCV),
