2、IF=1.693)AbstractPestedespetitsruminants(PPR),ahighlycontagiousdiseaseinducedbyPPRvirus(PPRV),affectssheepandgoats.PPRVfusion(F)proteinisimportantfortheinductionofimmuneresponsesagainstPPRV.WeconstructedaSemlikiForestvirus(SFV)replicon-vectoredDNAvaccine(“suic
3、idalDNAvaccine”)andevaluateditsimmunogenicityinBALB/cmice.ThepSCA1/FexpressedFproteininvitroandinducedspecificandneutralizingantibodyproduction,andlymphocyteproliferationinmice.MicevaccinatedwithpSCA1/FhadincreasedIL-2andIL-10levelsafter24-hpostfirstimmunizati
4、on.IFN-γandTNF-αlevelsincreasedfromthattimepointandgraduallydecreasedthereafter.Thus,theSemlikiForestvirusreplicon-vectoredDNAvaccineexpressingtheFproteinofPPRVinducedbothhumoralandcell-mediatedimmuneresponsesinmice.Thiscouldbeconsideredasanovelstrategyforvacc
5、inedevelopmentagainstPPR.1IntroductionPestedespetitsruminants(PPR;alsoknownasovinerinder-pest)isanacutediseasecausedbythePPRvirus(PPRV);PPRisaclassifieddiseaselistedbytheWorldOrganizationforAnimalHealth(OIE),anditischaracterizedbyhighfever,enteritis,pneumonia,
6、diarrhea,andnecroticinflammationwithseveremorbidityandhighmortality.Thehomologouslive-attenuatedvaccineNigeria75/1(Nig75/1)waspreviouslyusedtomitigatePPRV;however,itsusewasconfoundedbyissueswithdifferentiatinginfectedfromvaccinatedanimals(DIVA).Moreover,usageo
7、ftheheatsensitivePPRVvaccinesinhightemperatureareasisamajorconcern.Therefore,improvedvaccinesandDIVAmethodsforserosurveillanceandseromonitoringofPPRVareurgentlyneeded.Recently,suicidalDNAvaccineshaveemergedasanimportantstrategyforvaccinedevelopment,andarecapab
