4、oC-terminaltransmembranedomainsandhavebeencharacterizedasmagnesiumtransporters.Usingtherecentlyestablishedmag-fura-2systemallowingdirectmeasurementofMg2+uptakeintomitochondriaofSaccharomycescerevisiae,wefindthatallmembersoftheArabidopsisfamilycomplementthecorrespondingyeastmrs2mutant.Highlydifferen

5、tpatternsoftissue-specificexpressionwereobservedfortheMRS2/MGTfamilymembersinplanta.Sixofthemareexpressedinroottissues,indicatingapossibleinvolvementinplantmagnesiumsupplyanddistributionafteruptakefromthesoilsubstrate.HomozygousT-DNAinsertionknockoutlineswereobtainedforfourmembersoftheMRS2/MGTgenef

6、amily.Astrong,magnesium-dependentphenotypeofgrowthretardationwasfoundformrs2-7whenMg2+concentrationswereloweredto50mMinhydroponiccultures.EctopicoverexpressionofMRS2-7fromthecauliflowermosaicvirus35Spromoterresultsincomplementationandincreasedbiomassaccumulation.Greenfluorescentproteinreportergenefu

7、sionsindicatealocationofMRS2-7intheendomembranesystem.Hence,contrarytowhatisfrequentlyfoundinanalysesofplantgenefamilies,asinglegenefamilymemberknockoutresultsinastrong,environmentallydependentphenotype.INTRODUCT
