2、gust1988)Thenucleotidesequenceofthe3’-endofthegenomicRNAofhumancoronavirus0C43(HCV-0C43)wasdeterminedfromthecDNAclonesoftheintracellularvirus-specificmRNAs.Thenucleotidesequenceandthepredictedaminoacidsequenceofthemainopenreadingframe(ORF),whichrepresentsthenucleocapsid(N)pro
3、tein,werehighlyhomologoustothoseofbovinecoronavirus(BCV)Mebusstrain.ThisORFpredictsaproteinof448aminoacids.AdditionalsmallerORFsarealsopresentinadifferentreadingframe.Wehavealsodeterminedtheleadersequencepresentatthe5’-endofHCV-OC43mRNAsbyaprimerextensionstudy.Thissequenceish
4、ighlyhomologoustothatofmousehepatitisvirus,particularlyinthe3’-endoftheleadersequence,whichispostulatedtobeinvolvedintheuniquemechanismofleader-primedtranscription.ThesedatasuggestthatHCV-0C43andBCVmighthavedivergedfromeachotherfairlyrecentlyandthatthe3’-endoftheleadersequenc
5、ehassignificantfunctionalroles.Humancoronavirus;Nucleocapsidsequence;LeaderRNAHumancoronaviruses(HCV)areimportanthumanrespiratorypathogens,re-sponsibleforalmost20%ofcommoncoldsinwinterseasons(McIntosh,1974).Theyhavealsobeenassociatedwithdiarrheaofunknownetiology,suchasinfanti
