6、······················································································46IL-1β对大鼠终板软骨细胞增殖的影响及机制研究摘要目的:相间盘退变为多种因素综合影响所致,有报道表明,软骨终板发生钙化所引发椎间盘营养物质供应下降为椎间盘启动的关键性因素。本实验通过培养大鼠椎体终板软骨细胞,检测ADAMTS-5蛋白的表达水平与IL-1β是否具有相关性,在IL-1β炎性因子的干预下,通过检测磷酸化p65蛋白,p65蛋白含量的变化,来探究IL-1β是否促进ADAMTS-5的

7、表达通过NF-κB信号通路,从而促进终板软骨细胞退变,为预防和治疗椎间盘退变提供新的方法。方法:取4-6周龄大鼠椎体的终板软骨细胞进行纯化,通过原代及传代培养。选择生长较好的3代细胞进行实验,通过观察细胞的形态,来描绘细胞的生长曲线。1.采用MTT法研究不同浓度的IL-1β对大鼠终板软骨细胞增殖的影响。通过在紫外分光光度计上测定各孔光吸收值,以时间为横坐标,吸光值为纵坐标,观察细胞增殖变化。2.研究不同浓度的IL-1β(5ng/ml,10ng/ml、20ng/ml、40ng/ml)对大鼠终板软骨细胞ADAMTS-5蛋白表达的影响。3.培养皿中加入10ng/mL的
