1、华中科技大学硕士学位论文AbstractObjective:ToexploretheeffectsandpossiblemechanismofrecombinanthumanNeuregulin-1βoncardiacfunctionofacutemyocardialinfarction(AMI)rats,theangiogenesisandtheexpressionofvascularendothelialgrowthfactor(VEGF)inischemicmyocardium.Methods:AMImodelwasestablishedwithSprague-Dawleyr
2、atsbyligationoftheleftanteriordescendingcoronaryartery.Theratswhichsurvived24hoursaftersurgerywererandomlyseparatedinto2groups:rhNrg-1βtreatmentgroupwithcaudal veininjectionofrhNrg-1βatadoseofl0µg·kg-1·d–1for5consecutivedaysandthemyocardialinfarction(MI)controlgroup.Anothershamoperationgroupun
3、derwentthoracicincisionwithoutligationofleftanteriordescendingcoronaryartery.MIcontrolgroupandshamoperationgroupweresimultaneouslyadministeredwith0.1mol/LPBS.Afterroutinefeedandwaterfor28days,cardiacfunctionofdifferentgroupswereevaluatedbyechocardiography.Afterthat,theratsweresoonexecutedtodet
4、ecttheweightandmassindexofleftventricular.DensitiesofnewcollateralmicrovesselweremeasuredbyⅧfactorimmunohistochemicalanalysis.WesternblottingwasadoptedtoobserveVEGFexpressionintheischemicmyocardiumofAMIrats.Results:ComparedwiththeMIcontrolgroup,cardiacfunctionoftherhNrg-1βtreatmentgroupobvious
5、lyimproved;Comparedwiththeshamoperationgroup,alargenumberofnewbloodvesselsintheproliferativestagecanbefoundintheinfarctionborderzoneofbothMIgroupandrhNrg-1βtreatmentgroup,andcapillarydensityintherhNrg-1βtreatmentgroupwassignificantlygreaterthanthatintheMIcontrolgroup;Besides,theexpressionofVEG
6、FproteinsignificantlyincreasedintheinfarctionborderzoneofMIgroupandrhNrg-1βtreatmentgroup,andexpressionlevelintheMIgroupwasmuchlessthanthatintherhNrg-1βtreatmentgroup.Conclusions:rhNrg-1βincreaseexpressionofVEGFproteininischemicmyocardiumofAMIratswhichcanpromoteangiogenesisandcollateralcircula
