2、pulationofcelltypes.BMMNCswhichincludemultiplestemcellsandcandifferentiateintovesselwallcellsinthelimbischemiaandmyocardialinfarctioncanstronglypromoteneovasculogenesis.ThisstudyistoobservewhethertheBMMNCscanpromoteangiogenesisandarteriogenesisbydifferentiatingintoendothelialcellsandsmoothmusclecel

3、lsinaratmodelofischemicstrokeandwhetherBMMNCsexertlong—termneuroprotection.Materiaisandmethods1.Thetracer,isolation,identificationofcellularcomponentsandLabelingrateofBMMNCs.WeinjectedadultmaleSprague—DawleyratsintraperitoneallywithBrdU(50mg/kg/day)oncedailyfor14daysbeforeharvestingthebonemarrow;BM

4、MNCswereisolatedwithdensitygradientcentrifugationmethod;ThecharacterizationofCD34,CD45,CD90,CDl17andBrdUlabelingrateofBMMNCswereidentifiedwithflowcytometry.2.ExperimentalAnimalsandGroupsIVAdultmaleSprague-Dawleyrats(300-3109,3monthsold)wererandomlydividedintofourgroups:sham-operatedgroup(n=26),untr

5、eatedischemicgroup(n=34),vehicle—treatedischemicgroup(n=36),andBMMNC-treatedischemicgroup(n=54).3.Themodelofcerebralischemia,Celltransplantation,PreparationofsamplesandObservationindicatorThepMCAOmodelwasestablishedwithsutureembolimethod;BMMNCswereisolatedwithdensitygradientcentrifugationmethod.Bod

6、yweightchangeswereassessedweeklythroughday42afterstroke.TherecoveryofneurologicalfunctionaldeficitswereassessedasthesamedaysbymodifiedNeurologicalSeverityScores(mNSS).Infarctvolumewasassessedonweek1with2%2,3,5一triphenyltetrazolium-chloride/saline叮C)staining.Thediameterandmorphologicalcharacteristic
