1、浙江大学硕士学位论文格列齐特的合成姓名：梅光明申请学位级别：硕士专业：化学工程指导教师：张未星20050401浙江大学硕士学位论文摘要格列齐特(Gliclazide)是第二代磺酰脲类口服降糖药，具有降血糖和改善凝血功能的双重作用，不仅可以改善糖尿病患者的代谢，而且可以改善或延缓糖尿病JIⅢ管并发症的发生。本文在综述文献合成方法基础上寻找一条适合工业化生产的合成路线。本论文研究以环戊烷邻二甲酰亚胺为原料经过还原、弧硝化、还原反应，再与对甲苯磺酰脲缩合反应合成格列齐特的路线，重点考察了各步反应巾各影响因素对产物收率的影响。存酰业胺还

2、原反应中，用KBH4代替“AJH4，用ZnCl2代替A1CL3，甲苯和THF代替THF，即降低了成本，又减少环境污染，。重点考察了该还原反应中的甲苯和THF投料比，钾硼氢和氯化锌的比例，反应时间和温度及反应产物提纯等的影响，优化各步反应条件，收率达85％。在氮杂环亚硝化反应中，重点考察了反应中的温度、pH及浓度的影响，反应几乎等虽进行，收率达到99．5％。在亚硝基化合物的还原反应中，用zn和HCI来代替LiAll44．又用NaOH沉淀ZnCl2，既降低了成本，又利于三废处理。重点考察．r还原剂及酸的选择，使其适用于工业化生产。收
