1、不同培养条件对铁皮石斛类原球茎生物反应器培养的影响1,222113*徐步青,李振中,张俊,王芬,刘幸佳,崔永一1.浙江农林大学园林学院,浙江临安3113002.浙江农林大学林学基础省级实验教学示范中心,浙江临安3113003.浙江农林大学农业与食品科学学院,浙江临安311300摘要:目的研究不同培养条件对铁皮石斛类原球茎在生物反应器中培养的影响。方法采用接种量、通气量、蔗糖浓度、光照强度等单因素试验设计,烘干法测定生物量,苯酚-硫酸法测定多糖量,数据采用SPSS16.0软件分析。结果在生物反应器培养条件下,利于铁皮石斛类原球茎
2、增殖和多糖积累的培养基组分为:1/2MS基本培养基添加1.0mg/LNAA、5%椰乳、3%蔗糖,pH值为6.0。接种量为40g/L,采用孔径为15μm的多孔喷头,通气量用1.0L/min和0.5L/min交替使用以及光照强度为2000lx等培养条件有效地提高类原球茎增殖系数和多糖量。结论不同的培养条件对铁皮石斛类原球茎的生长、多糖的变化有显著影响,适宜的培养条件对铁皮石斛类原球茎的生物量和主要药用成分的生产具有重要的实践意义。关键词:铁皮石斛;类原球茎;培养条件;生物反应器;增殖系数;多糖中图分类号:R282.21文献标志码:
