1、华中科技大学同济医学院硕士学位论文细胞周期蛋白E及其依赖性激酶2在子宫平滑肌肿瘤的表达及临床意义华中科技大学同济医学院附属同济医院妇产科研究生张素丽导师郑红兵教授中文摘要目的:研究细胞周期蛋白E(cyclinE)及其依赖性激酶2(CDK2)在子宫平滑肌肿瘤(USMTs)中的表达情况及其与子宫平滑肌细胞增殖间的关系,以探讨其在子宫平滑肌肿瘤发病机制中的作用及临床意义。方法:1.采用免疫组织化学链霉菌抗生物素蛋白过氧化物霉(S—P)法分别检测cyclinE、CDK2和增殖细胞核抗原(PCNA)在20例正常子宫平滑肌组织(M),35例良性子宫平滑肌瘤(uL),18例子宫平滑肌肉
2、瘤(LMS)及22例交界性子宫平滑肌瘤(BLM):包括13例富于细胞型子宫平滑肌瘤(CL)、4例奇异型子宫平滑肌瘤(BL)及5例核分裂活跃型子宫平滑肌瘤(ML)的蛋白表达,并应用图像分析系统分别测定cyclinE、CDK2和PCNA阳性染色面积及平均光度;2.采用半定量RT—PCR方法分别检测cyclinE、CDK2和PCNA在良性子宫平滑肌瘤(UL)及正常子宫平滑肌组织(M)中的mRNA表达。结果:1.CyclinE、CDK2及PCNA在uL组、BLM组(包括CL组、BL组、ML组)和LMS组的蛋白表达均明显高于M组,差异具有统计学意义,P3、均明显高于uL组及BLM组,差异亦具有统计学意义,PO.05:CL组、BL组和ML组组问比较差异亦无统计学意义,P>O.05。2.CyclinE、CDK2及PCNA在UL组的mRNA表达亦高于M组,差异具有统计学意义,P4、DK2与PCNA之间亦呈正相关,相关系数分别为:r=O.623,P5、artmentofGynecologyandObstetrics,Tongjihospital,TongjiMedicalCollege,HuazhongScienceandTechnologyUniversityPostgraduate:ZhangSuliTutor:ProfessorZhengHongbingAbstractObjective:WemadeastudyoftheexpressionofcyclinEandCDK2inuterinesmoothmuscletumors(USMTs)andtherelationbetweencyclinEandCDK2an6、dtheproliferationofuterinesmoothmuscle,andinvestigatedtheeffectofcyclinEandCDK2inthegrowthanddevelopmentofuterinesmoothmuscletumors(USMTs)andtheirclinicsignificance.Methods:I.Immunohistochemicaltechnique(S-Pmethod)wasusedtodetecttheproteinexpressionofcyclinE,CDK2andPCNAin20normalmyometriu7、m(M),35usualIeiomyoma(UL)。181eiomyosarcoma(LMS)and22borderline1eiomyoma(BLM):Including13cellular1eiomyoma(CL),4bizarreieiomyoma(BL)and5mitoticallyactiveleiomyoma(ML).Thenweappliedtheimage—analysiS-systemtodetectthepositiveareaofcolourationandtheaverageluminosityofcy
