3、althoughinterdependent,brainregions.Theseincludeacerebralcortexcontainingprogenitorpopulationsthatorganizeandproducematurecorticalneuronsubtypes.Furthermore,cerebralorganoidsareshowntorecapitulatefeaturesofhumancorticaldevelopment,namelycharacteristicprogenitorzoneorganizati

4、onwithabundantouterradialglialstemcells.Finally,weuseRNAinterferenceandpatient-specificinducedpluripotentstemcellstomodelmicrocephaly,adisorderthathasbeendifficulttorecapitulateinmice.Wedemonstrateprematureneuronaldifferentiationinpatientorganoids,adefectthatcouldhelptoexpla

5、inthediseasephenotype.Together,thesedatashowthatthree-dimensionalorganoidscanrecapitulatedevelopmentanddiseaseeveninthismostcomplexhumantissue.Mammalianbraindevelopmentbeginswiththeexpansionoftheneuro-ofregionalidentitiesorganizedasdiscretedomainscapableofinfluen-1epithelium

6、togenerateradialglialstemcells(RGs).TheseRGsdivideatcingoneanother.Furthermore,cerebralcorticalregionsdisplayanorgani-theapicalsurfacewithintheventricularzone(VZ)togenerateneuronszationsimilartothedevelopinghumanbrainatearlystages,aswellasandintermediateprogenitors.Intermedi
