2、ationofclinicalsamplesforevidenceofinfection.Duringtheepidemicofsevereacuterespiratorysyn-drome(SARS)in2003,PCRnotonlyofferedarapidwaytodiagnoseSARS-coronavirus(SARS-CoV)infection,butalsomadethemolecularanalysisofitsgenomicsequencepossible.SequencevariationswereobservedintheS

3、AR-CoVobtainedfromdifferentpatientsinthisepidemic.Theseuniqueviralgeneticsignaturescanbeappliedasapow-erfulmoleculartoolintracingtherouteoftransmissionandinstudyingthegenomeevolutionofSARS-CoV.ToextractthiswealthofinformationfromthelimitedprimaryclinicalspecimensofSARSpatient

4、s,wewerepresentedwiththechallengeofefficientlyamplifyingfragmentsoftheSARS-CoVgenomeforanalysis.Inthischapter,wewilldiscusshowwemanagedtoaccomplishthistaskwithouroptimizedprotocolsonreverse-transcription,nestedPCRamplification,andDNAcyclesequencing.Wewillalsodis-cussthesequen
