3、nservedmotifspresentintheknownNBS-LRRR-genesequencesandRgeneanalogs(RGAs),severaldegenerateprimersweredesignedandappliedintheRGAisolationfromErianthusarundinaceususingPCRapproach.Intotal,6RGAsweresuccessfullyobtained,withGenBankaccessionnumbersofEU685835,EU685836,EU68
4、5837,EU685838,EU685839,andEU685840.MultiplealignmentsshowedthattheencodingsequencesofthesixcloneswerehighlyconservedandstrikinglysimilartotheelevenmosttypicalNBS-LRRtypeR-genepeptidesequences,especiallyatthefourNBSmotifsofP-loop,kinase-2,kinase-3a,andHD.Theidentityper
5、centageattheamino-acidlevelrangedfrom8.3%to93.0%amongall17sequencestestedandfrom30.5%to45.6%amongthesixRGAsclonedinthisstudy.Theresultsofclusteranalysisandtheexistenceofanasparticacidresidue(D)atthefinalresiduepositionofthekinase-2motifalsoindicatedthatallofE.arundina
6、ceusRGAsmightbelongtonon-TIRgroup.TheReal-timePCRresultsshowedthatalloftheRGAswereconstitutivelyexpressedinroots,stalksandleavesofE.arundinaceus,andtheirexpressioncouldbeup-regulatedinleavesbytheexogenoussignalmoleculesSAandH2O2.Therefore,itsuggestedthatE.arundinaceus
