1、摘要本论文以欧洲花楸带芽茎段、无菌叶片和复叶轴为外植体,研究了欧洲花楸组织培养技术体系的建立,主要研究内容和结果如下:(1)外植体的消毒效果差异较大,春季带芽茎段以饱和次氯酸钙溶液消毒15rain最佳,而夏季带芽茎段则需要30rain,但消毒效果仍较差,夏季带芽茎段用0.1%升汞消毒处理要好于饱和次氯酸钙溶液,最佳时间为8min。总体来看,春季带芽茎段比夏季带芽茎段易于消毒。(2)带芽茎段萌发的最佳培养基为MS+6.BA2.0mg/L+NAA0.1mg/L,春季带芽茎段和夏季带芽茎段的萌发率分别为89.
2、47%和64.00%。(3)再生苗的继代增殖培养基为1/2MS+6一BAl.0mg/L+NAA0.2mg/L。(4)叶片诱导愈伤组织的最佳培养基为MS+6一BA2.0mg/L+NAA0.1mg/L或MS+6-BAt.0mg/L+IBA0.05mg/L,复叶轴诱导愈伤组织的最佳培养基为MS+6-BA2.0mg/L+NAA0.05-0.1mg/L或MS+6-BA2.0mg/t.+mA0.05mg/L,经诱导产生的愈伤组织生长状态好,增殖快,但未经过分化而产生不定芽:暗培养对叶片和复叶轴愈伤组织的诱导与分化没
3、有影响。(5)复叶诱导不定芽的最佳培养基为MS+6·BA2.0mg/L+KTI.5mg/L+NAA0.05m班,复叶轴诱导不定芽的最佳培养基为MS+6·BAl.0mg/L+KTI.5mg/L+NAA0.05m班,在这两种培养基上诱导分化的芽生长健壮,芽多而高,经过增殖培养后能诱导根的生成。暗培养有利于复叶和复叶轴诱导不定芽,能提高其分化率及每个复叶和复叶轴分化的芽数,最佳的暗培养时间为三周。(6)欧洲花楸在I/2MS+IBA0.01mg/L或I/2MS+2,4-DO.03mg,L的培养基上,生根率最高,
