1、骨髓间充质干细胞对失血性休克大鼠早期炎性反应的影响作者:李彤王子薇王淑梅张学军李艳秋单位:吉林大学口腔医院通讯作者:王淑梅单位:吉林大学第二医院麻醉科基金项目:吉林省产业技术研究与开发项目吉发改高技【2013】779号【摘要】目的探讨骨髓间充质干细胞对失血性休克大鼠血清炎性因子及重要脏器的影响。方法体外分离培养大鼠骨髓间充质干细胞。雄性SD大鼠随机分为假手术组、模型组、治疗组(n=20)。改良Wiggers法制备大鼠出血性休克模型,休克持续90min。治疗组在复苏前给予106个第3代骨髓间充质干细胞PBS悬液1ml,假手术组和模型组给予相同体积PBS溶液。分别
2、于复苏后6h、12h、24h、48h、72h取血清,ELISA检测TNF-α,IL-1β、IL-6、IL-10水平,HE染色观察心、肝、肺、肾病理损伤。结果治疗组血清TNF-α和IL-6在复苏后12h-72h内较模型组显著下降(p<0.05);治疗组血清IL-1β在复苏后6h-12h内较模型组下降显著(p<0.05);治疗组IL-10水平在复苏后6h-48h内均较模型组有显著升高(p<0.05)。骨髓间充质干细胞减轻肺、肝、肾、心等器官的炎性细胞浸润。结论骨髓间充质干细胞能调节失血性休克早期重要炎性因子水平,促进机体促炎-抗炎系统平衡。【关键词】骨髓间充质干细
