2、engthened.Asanimportantentomopathogenicfungus,Nomuraearileyiiscapableofinfectingseverallepidopterouspests,causinginsectdiseaseinnaturalenvironment.Asanimportantbio-controlagent,theeffectivecomponentofN.rileyiisconidia,whichrequirefastidiousgrowthconditionsalongwith
3、stimulatorylight,thus,limitingitsmassproductionandcommercialization.OurlaboratorysuccessfullyaccomplishedN.rileyimicrosclerotium(MS)productioninliquidamendedmedium(AM),whichexhibitedinsecticidalefficacyonSpodopteralitura,andconfirmedthatMScanbeusedasanalternativeac
4、tiveagenttoconidiainpestbio-control.Subsequently,throughtranscriptionanalysis,wefoundthatMSdifferentiationassociatedwithoxidativestressandpolarizedgrowthofhyphae.Inthepresentstudy,weclonedtheracAandcdc42genesthatwereupregulatedduringMSdifferentiationandcharacterize
5、dtheirinvolvementinMSdevelopmentandpolarizedgrowth.Theresultsofmainresearchareasfollow:①Cloningandidentificationofthetargetgenes:Thefull-lengthcDNAsequencesoftheN.rileyiracAandcdc42geneswereobtainedbySMARTRACERT-PCR.TheputativecodingsequencesofracAandcdc42were657an
6、d555bp,encoding218and184aminoacidresidues,respectively.②Bioinformaticsanalysisofthetargetgenes:ProteinsequenceanalysisfoundthatbothRacAandCdc42harborthetypicalfiveGTP/GDP-bindingorhydrolysismotifs(G1–G5)andtwomolecularswitchescharacteristicoftheRho-familysmallGTPas
7、e;ThephylogenetictreeanalysisdemonstratedthattheRacAorCdc42proteinsofN.rileyiarehighlyhomologoustothoseofMetarhiziumacridum.③ThedynamicprocessofMSdevelopment:WeobservedthedynamicprocessofMSformationwithmicroscopeandfoundthatitwascomposedofwindingpolarhyphaeandwasac
