2、ousticspectrummonitor,toanalyzeflorasimilarity,thenumberofEscherichiacoli,lactobacillus,methanogens,andconcentrationdistributionofC02,CO,N20,NH3,CH4,H,Oindifferentdepthofbio-bedforthefurtherstudyofcommunityecologyonbio-bed.DuetothecomplexcompositionsofthelitterinBio-
3、bed,therearemanyimpuritiesinDNAex臼?act.Underthesecircumstances,theuniversallyapplicableandreproduciblemetllodtoextractDNAfromdifferentorganismsisnotdesirable.Thus,theaimofthisstudywastomodifytheconventionalmethodinordertoextracthighqualitycomplex2enomicDNA行omdifferen
4、tbacteriainBio.bed.OurresultsdemonstratedthatmodifiedGenoiseBacteriaDNAEctractionkitCanremovePCRinhibitingfactoreffectively,andtheoptimalconcentrationofttliaminehydrochloridewasO.5M.Throughthetestofbacterialuniversalprimer968f/1401r,bacterial16SrDNAsequences.whichwer
5、esamplesfromthelitterinbio—bedofTaicangpigbreedingfarm,wereamplifiedbyPCR,andusingtheDGGEfingerprinttechnologytoanalysetheproductsofthePCR.ItWasfoundthatthenumberofbacterialbandsinahigherlevel(O~15cm)oflitterinbio.bedwassimilarandthefluctuationrangewasfrom24to27.Then
6、umberofbacterialbandsinthedepthof20cmwasthelowest,whilethenumberinthedepthof30cm陀achedthemaximum,Thesimilaritybetweenthedepthof10cmandotherdepthsoflitterinbio-bedWasthelowest,only51.2%,followedWasthesurfacelayerofpadding,andthesimilarityofresiduallevelsWas56.8%;thesi
7、milarityofthesurfaceandthedepthof5cmWastheclosest,reaching95.6%.ByReal.TimePCRtechnology,thispapermadeaquantitativeanalysisofE.coli,LactobacilluSandMethanogensoflitterindifferentdepthsonbio-bed.ThestudyfoundthatthelargestnumberofE.coliwasonthesurfaceofbio-bed,whichWa
