1、AbstractABSTRACTObjective:ToinvestigatethedifferenceofGFAP、VEGF、Bcl-2、BaxandTUNELpositivecellsintheischemiapenumbra,thelevelofTNF-inserum,thescoreofmNSS,thecountofbodyweightandthedistributionofstemcellsinorgansbetweenintravenousandintracarotidrouteforbonemarrowderivedmesenchymalstemcellsinth

2、etreatmentofMCAO6hours.ToinvestigatethedifferenceofintravenousandintracarotiddeliveryrouteandthemechanismofBMSCsinthetreatmentofMCAOMethods:1.Themiddlecerebralarteryocclusionmodelwasconductedbymodifiedintraluminalsuturemethod.2.Thebonemarrowderivedmesenchymalstemcellswereisolated,purifiedand

3、culturedbywholebonemarrowadherentcultureandidentifiedbyflowcytometry.BMSCsweredyedwithCM-dilfortracing.3.TransplantionwasoperatedatMCAO6hoursbyintravenousandintracarotidrouterespectively.4.7daysaftertransplantation,thescoreofmNSSandthecountofbodyweightwereevaluated.5.ThelevelofTNF-αinserumwa

4、sdetectedbyELISA.6.TheexpressionofGFAP、VEGF、Bcl-2andBaxweredetectedbyimmunohistochemistry.7.CellapoptosiswasdetectedbyTUNELstaining.Results:1.TheexpressionofsurfaceantigenCD90、CD29、CD106、CD11b、CD34、CD45wererespectively95.7%、97.3%、52.7%、6.01%、2.95%、2.26%.2.ComparedwithPBSgroup,thescoresofmNSS

5、wassignificantlydecreasedintheBMSCsgroups,thedifferencebetweenintracarotidandintravenousBMSCsgroupwasnotshowed.ComparedwithintracarotidPBSgroup,thecountofbodyVAbstractweightwasincreasedintheintracarotidBMSCsgroup,andtheresultwasthesamebetweentheintravenousPBSandBMSCsgroup.Comparedwithintrave

6、nousPBSgroup,thedifferencewasnotshowintheintracarotidBMSCsgroup.ThedifferencebetweenintracarotidandintravenousBMSCsgroupwasnotshowed.3.TheGFAPpositivecellswereobservedintheischemiapenumbrasurroundingtheischemiacoremainly.Regardlessthedeliveryroute,comparedwithPBSgroup,thenumberofGFAPpositive
