3、ptedJuly9,2012;advancepublicationreleasedonlineAugust3,2012Todevelopastablechronicobstructivepulmonarydisease(COPD)modelinrats.Sprague-Dawleyratsweretreatedwithcigarette-smokeinhalation(CSI)for12weeks,repetitivebacterialinfection(RBI)for8weeks,orthecombinationofthe

4、two(CCR)for12weeksandfollowedupfortheadditional20weeks.Tidalvolume(VT),peakexpiratoryflow(PEF)and50%VTexpiratoryflow(EF50),histologicalchangesinthelungs,andlevelsofthecytokinestumornecrosisfactor(TNF)-α,interleukin(IL)-8,andIL-10inserumandbronchialalveolarlavagefluid(

5、BALF)wereexaminedatintervalsduringthe32weekstudyperiod.Therightventricularhypertrophyindex(RVHI)wasalsodeterminedatthesametimes.VT,PEF,andEF50weredecreasedinratswithCOPDcomparedtothecontrol.TheexpressionofTNF-α,IL-8andIL-10increasedinbothserumandBALFwithasimilartre

6、nd.BronchioleandarteriolewallthicknessandthedegreeofbronchiolestenosisandalveolarsizeincreasedinCOPDrats.RVHIwasreducedgraduallyfollowingthetreatment.AllofthesechangesweremorepronouncedintheCCR-treatmentgroupthanintheothergroups.OurresultshaveshownthatCSIorRBIalone

7、caninduceCOPDinrats,butthatthecombinationofCSIwithRBIinducesastableCOPDthathasmoresimilaritytocomplicationsseeninpatientswithCOPD.ThiscombinationmaythereforeprovideamoreappropriatemodelforstudyofhumanCOPD.Keywordspulmonarydisease;chronicobstructive;animalmodel;pulm
