2、robiology,UniversityofSouthernCalifornia,SchoolofMedicine,LosAngeles,California90033Abstract.MonoclonalantibodiesreactingwiththeA59strainofmousehepatitisvirus(MHV-A59)werecharacterizedandthosespecifictotheE2majorenvelopeglycoproteinwerestudiedindetail.Antibodiesweretestedfortheirab
3、ilitytoneutralizeviralinfectivity(N'characteristic)andpreventviral-inducedcell-to-cellfusion(F'characteristic).AllfourpossiblecombinationsofactivitiesreflectingE2functionswerefound,i.e.,N+F+,N-F-,N'F-,andN-F+.Inaddition,competitivebindingstudieswiththesemonoclonalantibodiesrevealed
4、twononoverlappingan-tigenicregions.Thefirstregion,designatedA,wasrecognizedbyantibodieswhichincludedeachofthefourfunctionaltypes.RegionBwasidentifiedbyasinglemonoclonalantibodywithN-F-activities.Theexistenceofantibodieswhichonlyneutralizevirusoronlyblockviral-inducedfusionimpliesth
5、atthestructuresonE2whichserveastargetsforneutralizationandwhichinducefusionarenotidentical.ThecriticaldeterminantsforneutralizationandfusionmustbecloselyrelatedtopographicallyonE2sincebothN+F-andN-Ffantibodiesrecognizethesameantigenicregion.01987SocietyforExperimentalBiologyandMedi
