1、摘要本研究根据Calnek疫苗株序列设计了3l条引物,利用反转录聚合酶链式反应,对我国AEV分离株kZ株的全基因组核苷酸序列进行了测定,同时,采用Bac-to-Bac系统在昆虫St9细胞中成功表达了AEVVPI基因,现将结果报告如下:对我国AEV分离株kZ株的全基因组核营酸序列测定的结果表明:该病毒株基因组全长为7059个核苷酸残基,包括494个核苷酸残基的5’非编码区、6402个核苷酸残基的开放阅读框和136个核苷酸残基的3’非编码区及耐y(A)尾巴。与已发表的AEV疫苗株l143的基因组序列比较发现,它们之间核苷酸和氨基酸的同源性分别为98%和97.6%,与鸡胚
2、适应株VR的基因组序列比较核苷酸和氨基酸的同源性分别为93.9%和96.4%。结构蛋I皇I(VPl-VP4)中,主要宿主保护性免疫原蛋白VPl氨基酸之间差异较小,kZ株与疫苗株和鸡胚适应株VPl基因的氨基酸同源性分别为99.3%和98.5%。重组Ba蕊nid-VPl转染昆虫细胞SD后,获得的重组病毒,经SDS-PAGE可见表达的AEVVPl融合蛋白,大小约为35kD。经蛋白纯化用Westernblot检测,初步证实表达的AEVVPl蛋白具有抗原活性。关键词:禽脑脊髓炎病毒;全基因组序列测定;Bac.to-Bac系统:VPI基因表达Abstract31ofprimer
